ABSTRACT
Developing an Agrobacterium-based transformation system for chickpea involves identifying conditions to induce
Agrobacterium to transform without subsequently interfering with the regeneration of transformed cells. T-DNA transfer efficiency was investigated using ²-glucuronidase as reporter gene. Early detection of transient gene expression by GUS activity was employed for optimizing various parameters affecting transformation in chickpea. Various parameters like effect of bacterial conc., pre-culturing of explants, effect of wounding, co-cultivation duration and effect of light were studied.
