This research was aimed at determining the effect of daun wungu [Graptophyllum pictum (L.) Griff] ethanol extract on testis histology of male mice induced by cadmium. The addition of cadmium sulphate (CdSO4) was intended to create oxidative stress. Twenty-five male mice used in this research were divided into five groups, namely, the control group (0.1ml aquadest), negative control [CdSO4 20 mg/kg body weight (bw) for 10 days and aquadest for 25 days], three groups treated with CdSO4 20 mg/kg bw for 10 days, then given ethanol extract of daun wungu with doses of 25, 50 and 75 mg/kg bw for 25 days, respectively. All mice after the end of the research were sacrificed, the pair of testis organ were cut off. The testis histology was made using the paraffin method and Haematoxylin and Eosin staining. The result of this research showed that administration of cadmium damaged the testis histology due to oxidative stress and the addition of daun wungu ethanol extract showed improvement in testis histology. The ethanol
extract of daun wungu contained flavonoid substance which functioned as antioxidant. The dose of 50 mg/kg ethanol extract of daun wungu affected the improvement of testis histology (the number of spermatogenic cells, epithelial thickness and seminiferous tubule diameter) induced by cadmium.
