Assessment of Genetic Relationship among Merremia spp. by RAPD Technique

ABSTRACT

Genetic relationship of Merremia spp. had a significant role in developing the genetic sources which are needed to breed plants related to habitat process and utilization. The purpose of this study was to find the genetic relationship of Merremia spp. based on DNA bands using RAPD technique and DNA bands that influenced the clustering of Merremia spp. into the dendrogram. The method used to analyze genetic relationship of Merremia spp. was by random amplified polymorphic DNA or RAPD. Three primers used were OPA 1, OPA 3 and OPA 18 which could produce DNA bands clearly and well-separately. The number of DNA bands from amplification process using those primers was 18 band patterns with the length between from 400 to 1000 bp. The emerged bands were translated into binary data and analyzed with MVSP program version 3.22. The conclusion of this study was that the closest genetic relationship was found in M. peltata and M. mammosa which yielded similarity index of 0.571 or 57.1%. Simultaneously, the least close genetic relationship was M. umbellate and M. peltata which scored similarity index of 0.357 or 35.7%. The result of principal component analysis (PCA) showed that the band length as 400, 650, 900,
1000, 1400 and 1600 bp from PCA had significant role in clustering Merremia spp. into the dendrogram.
